La Iglesia Ortodoxa de Antioquía llegó a Argentina con los primeros inmigrantes sirios y libaneses del siglo XIX que escapaban del Imperio Otomano. 

Al inicio, todos los ortodoxos porteños celebran juntos su culto en la Iglesia de la Santísima Trinidad en Parque Lezama. Luego, los ortodoxos antioquenos contaron con su propia catedral de los Santos Pedro y Pablo en Suipacha 846. Pero en la década del ‘40 la comunidad se trasladó a su actual ubicación. El proyecto fue realizado por el arquitecto Kirilos Nasif, en un estilo puramente oriental, adornando la Iglesia con íconos y vitrales. La primera liturgia se celebró el 24 de marzo de 1946.
